Cooperative mechanism in epigenetic network controls gene expression quantity.

摘要
Gene expression is controlled by signaling activities and epigenetic regulation. To understand quantitative mechanism of gene regulatory machinery, we analyze multi-omics data on histone modification, chromatin accessibility and single cell mRNA expression using a mathematical model. Our study using antigen-stimulated B cell indicated that multiple binding of NF-kB transcription factor on chromatin accessible region of the longer size of enhancer can induce higher level of gene expression than that is controlled by fewer binding of the NF-kB on the shorter size of enhancer. Our analysis indicates that the enhancer length is critical for quantitative control of gene expression. Imaging analysis further indicated that a cooperativity in transcription factor interaction arises from liquid-liquid phase separation in nucleus might be responsible for the event.
